I haven't had trouble with any of the bosses so far. The two chicks, i think i had a couple broadswords that I got from the entry way of one of the casinos.. thats one of the most powerful melee weapons.
One of the best weapons to get is the Wolverine like claws, thats all im using now.. Right from the main safe house walk towards the middle and look for that magazine stand, jump on the counter, jump to the roof of it, and theres a bowie knife, grab that and right by there is a sports store. get the boxing gloves and combo those two together. when you fight the bosses i try to have a sword, and the claws.. and two big health items, vodka, or oj. Then I just run up on them and mash the X button, i think most of the bosses you can hit twice, before they hit back. So hit twice and then step back, it also helps to have that dodge move, where u push the thumb stick in, so you roll outta the way.

bosses are kinda easy, the chef was the easiest, just get the guy up high on the roof of the resturants across from him. he'll help you beat em. the bowie and gloves are the best, and one gun atleast, mix voka and vokda or beer and beer in the blenders. it makes a painkiller and lets you go sav without takin much damage. the bitches were easy too. just smash on one of them and the other will off herself
any bar at a casino should have one, check the food court also.
as for the mascot, i just beat him on first try, run upstairs to the grocery mart and throw oil on the floor, everytime he rides over he'll slip, then do a spin with flame everywhere, after that you can get 2 good strikes on him with the bowie gloves. plus theres infinte pizza to eat.
